Walker, who was named England captain for Saturday's clash with Brazil, was substituted in the 20th minute after appearing to injure his hamstring while returning to stop a shot off the line from Vinícius Júnior early match.
“We do not know [how serious it is]”He hasn't had many injuries, so he doesn't know if it's just a strain or something more serious. We'll know more in the coming days.”
Maguire lasted until 67 minutes at Wembley on Saturday, before being replaced by Lewis Dunk.
The latest wave of withdrawals is a further blow to Southgate, who has already seen Harry Kane and Bukayo Saka withdraw from the England squad in recent days.
Kane, who was in the stands during England's defeat to Brazil on Saturday, had linked up with the team ahead of the match despite injuring his ankle in Bayern Munich's win over Darmstardt before the international break .
Speaking after Saturday's match, Southgate confirmed the striker would return to his club to recover from his injury and would not feature in England's friendly against Belgium on Tuesday.
Saka, who like Kane joined the England squad ahead of this round of friendlies, left the St. George's Park camp last week after the FA announced he had arrived with an injury and would was unable to participate in training sessions.
Manchester City defender Rico Lewis, who made his debut against North Macedonia in November, and uncapped Burnley goalkeeper James Trafford have been transferred from the under-21 team to the senior team.
